Could somebody tell me what is the difference between fitness first plus and fitness first, and the price difference between the two?

The plus ones have additional facilities like for example tennis courts at the Excelsior one but I think they are all different depending on the location. Probably best to visit the one you plan to go to and check it out for yourself - they do different deals every month and compare it to others in the area.
